There have been Sislers living in Newmarket and the surrounding area since 1807 and possibly earlier. The street was named Sisler Avenue because our family has lived in this area for so long. These Sislers are descendants of George Sisler who appears in the 1790 Pennsylvania census as living in Northumberland County. George actually came to live in Whitchurch Township in his later years.

Margaret Willson, eldest daughter of Joshua Willson, and Judith Ash, was born December 14th, 1786 at Beaver Harbour (now Pennfield), New Brunswick. Her parents, Loyalists, came in 1783 to what was known at that time as Nova Scotia. Her father, Joshua Willson was originally from Piscataway in New Jersey. The Willson family moved to Whitchurch Township, York County in 1811.
It was at Whitchurch that Margaret Willson met George Sisler/Sisley. Margaret was a Quaker and George applied for, and was accepted into, that faith at the Yonge St. location, before getting approval to marry her. They were married on February 18th, 1818.
Margaret and George Sisley are known to have had seven children who survived to adulthood, four sons and three daughters. Judith 1818, Joshua 1821, Bowley 1825, Margaret 1826, Catherine 1830, Paul 1832, and Robert 1833.
Margaret survived her husband George by some 52 years and died of old age in Richmond Hill at age 100 years 5 1/2 months 4 days.

The Quaker connection to the surname SISLER in Ontario seems to begin in 1817, when one George SISLER applies for membership in the Yonge Street Monthly Meeting, and is accepted into membership.
Shortly afterwards, on 18th of 2nd month 1818, George SISLER was married in a Quaker ceremony to Margaret WILLSON, and the record shows that he was the son of George SISLER and Margaret, who is deceased. At the time of the Separation of 1828, George SISLER, his wife Margaret and 4 children are identified with the Hicksite faction of the Pickering branch of the Yonge Street Meeting.
They were later disowned for non-attendance of meetings.
A gravestone for Margaret SISLEY (1786-1887) stands in the Presbyterian Cemetery at Richmond Hill (the surname seems to alternate between SISLER and SISLEY). There is no proof of a connection between this George SISLER and the James SISLERs Sr. and Jr. mentioned in the query, but the rare surname and the Quaker connection in both cases makes a relationship most likely.
Perhaps James SISLER Sr. and George SISLER Jr. were brothers, both children of George and Margaret SISLER.
